Rybelsus's elimination half-life is about 1 week, per the FDA label: that is how long it takes for the amount of oral semaglutide in your body to halve. Levels peak about 1 hour after a dose, and on the label schedule (a daily tablet) they reach steady state about 4 weeks after the first dose. After a last dose they fall below 5% of that steady level in about 4 weeks.
Between scheduled doses, levels swing between a peak and a trough of about 91% of that peak, which is why the chart looks like a sawtooth rather than a flat line. The label also reports that 0.4% to 1% of each dose is absorbed into circulation; the chart's mg figures build that in.
What if you miss a dose of Rybelsus?
The Rybelsus label says a missed dose should simply be skipped, with the next dose taken the following day.

That is why tablet doses run 3, 7, and 14 mg while injection doses run 0.25 to 2.4 mg: a 14 mg tablet actually delivers well under 1 mg of semaglutide into the body. Why is Rybelsus taken daily if its half-life is a week?
Because so little of each tablet is absorbed (0.4% to 1%, per the label), a small daily top-up is the practical way to keep enough semaglutide arriving. The long half-life then smooths those small daily additions into nearly flat levels.
Does a higher Rybelsus dose take longer to leave your system?
Not meaningfully. Rybelsus is eliminated proportionally: the amount cleared per day is a fixed fraction of what remains, so every dose falls along the same percentage curve on the same timeline. A higher dose means more oral semaglutide at every point, not a longer goodbye.
