Wegovy is semaglutide, with an elimination half-life of about 1 week and peak levels 1 to 3 days after a dose, per the FDA label. On the label schedule (a weekly injection), levels reach steady state about 5 weeks after the first dose, and after a last dose they fall below 5% of that steady level in about 5 weeks.

What the Wegovy label says about a missed dose

The Wegovy label uses a 48-hour rule: take a missed injection as soon as possible if the next scheduled dose is more than 2 days away, and skip it if less. For the daily tablet, the label says to skip a missed day and take the next dose the following day.

That window is the half-life doing the work: with about 1 weekof half-life, a late dose still lands on a meaningful base. This is the label's instruction to prescribers, not our advice; if you have missed a dose, your own label and prescriber are the authority.

One clock, three Wegovy forms

Wegovy now comes as the 2.4 mg weekly injection, the higher 7.2 mg Wegovy HD weekly injection, and the 25 mg daily tablet. All three are semaglutide, so all three share the one-week half-life and the weeks-long taper; what differs is the dosing rhythm and how much reaches your blood. The tablet is taken daily because of how the oral form absorbs (well under 1% of the swallowed mg), not because it leaves the body faster.

That has a counterintuitive consequence the calculator makes visible: stop the daily tablet and the accumulated semaglutide still takes about a month to fall below 5% of its steady level, nearly the same goodbye as the weekly injection.

Is it safe to restart Wegovy after a 3-week break?

That is your prescriber's call, and the label is the reference. A single missed injection follows the 2-day rule above; if 2 or more consecutive doses are missed, the Wegovy label says to reinitiate dose escalation at a lower dosage to reduce the risk of stomach side effects. Why does Wegovy feel weaker right before the next dose?

Two things stack. By the end of the week your level really is at its trough, about 62% of the post-dose peak. But blood level is not the same as felt effect: appetite suppression has its own timetable, and receptors adjust, so the day-6 dip many people report is real without meaning the medication is gone.

Does a higher Wegovy dose take longer to leave your system?

Not meaningfully. Wegovy is eliminated proportionally: the amount cleared per day is a fixed fraction of what remains, so every dose falls along the same percentage curve on the same timeline. A higher dose means more semaglutide at every point, not a longer goodbye.

Are these numbers my actual blood levels?
