How Long Does Foundayo Stay in Your System?

Foundayo's elimination half-life is 29 to 49 hours, per the FDA label: that is how long it takes for the amount of orforglipron in your body to halve. Levels peak 4 to 8 hours after a dose, and on the label schedule (a daily pill) they reach steady state about 8 days after the first dose. After a last dose they fall below 5% of that steady level in about 7 days.

Between scheduled doses, levels swing between a peak and a trough of about 72% of that peak, which is why the chart looks like a sawtooth rather than a flat line. The label also reports that 77% of each dose is absorbed into circulation; the chart's mg figures build that in.

The Foundayo label says to take a missed dose as soon as possible and not to double up the next one. If 7 or more doses in a row are missed, it instructs restarting dose escalation at a lower dose to reduce the risk of stomach and gut side effects.
